The joke is that the husband and his friend went to Thailand and his friend transitioned. Now the old friend is "missing" and the husband is married to who that friend became.

I understood "we met shortly after" as talking about the wife and the best friend. Like a hyperbolic expansion of the "adult men don't talk about their lives with their male friends."
